چکیده

Abstract Increasing evidence demonstrates that circular RNA (circRNA) plays a pivotal role in the development of disease, especially Cancer. A previous circRNA microarray study showed circGAK (hsa_circ_0005830) was remarkably down-regulated hepatocellular carcinoma (HCC) tissues. However, HCC remains largely unclear. The candidate circRNAs were screened via integrating Gene Expression Omnibus (GEO) database (GSE164803) analysis with online program GEO2R. Quantitative real-time PCR (qRT-PCR) employed to measure expression miR-1323, and hedgehog-interacting protein (HHIP) tissues cells. biological function examined using colony formation assay, 5-ethynyl-2'-deoxyuridine (EdU) wound healing transwell cell invasion endothelial tubular western blot xenograft mouse model. Bioinformatics analysis, immunoprecipitation (RIP) dual-luciferase reporter assay utilized test interaction between or HHIP. abnormally associated tumor-node-metastasis (TNM) stage. Overexpression impeded proliferation, migration, invasion, tube vitro, tumor growth vivo. predicted interacted which targeted HHIP mRNA 3'untranslated regions (UTR). Furthermore, upregulation miR-1323 shRNA-mediated suppression could recover circGAK-mediated malignant behaviors cells Taken together, circGAK/miR-1323/HHIP axis suppress progression may provide potential new targets for diagnosis therapy HCC.

The miR-383-LDHA axis regulates cell proliferation, invasion and glycolysis in hepatocellular cancer

Objective(s): To explore the correlation between expression patterns and functions of miR-383 and LDHA in hepatocellular cancer (HCC). Materials and Methods: We detected the expression of miR-383 and LDHA in 30 HCC tissues and their matched adjacent normal tissues using qRT-PCR. Then we performed MTT assay, foci formation assay, transwell migration assay, glucose uptake assay and lactate produc...

miR-1301 inhibits hepatocellular carcinoma cell migration, invasion, and angiogenesis by decreasing Wnt/β-catenin signaling through targeting BCL9

Metastasis is the major cause of the poor prognosis of hepatocellular carcinoma (HCC), and increasing evidence supports the contribution of miRNAs to cancer progression. However, the exact relationship between the level of miR-1301 expression and HCC cell migration, invasion, and angiogenesis remains largely unknown. Quantitative PCR was used to evaluate the level of miR-1301 expression in HCC ...

MiR-132 inhibits cell proliferation, invasion and migration of hepatocellular carcinoma by targeting PIK3R3.

MicroRNA-132 (miR‑132) has been reported to play a tumor suppressive role in different human malignancies. However, its role and underling mechanism in hepatocellular carcinoma (HCC) remains poorly defined due to lack of target gene information. In the present study, we demonstrated that the mean level of miR‑132 in hepatocellular carcinoma (HCC) tissues was significantly lower than that in mat...

MiR-26b inhibits hepatocellular carcinoma cell proliferation, migration, and invasion by targeting EphA2.

Deregulated microRNAs (miRNAs) have been shown to play important roles in cancer progression as a result of changes in expression of their target genes. In this study, we investigated the expression of miR-16b in eight hepatocellular carcinoma (HCC) cell lines, revealed the roles of miR-26b on hepatocellular carcinoma (HCC) cell proliferation, migration, and invasion, and confirmed that EphA2 i...

miR-940 Suppresses Tumor Cell Invasion and Migration via Regulation of CXCR2 in Hepatocellular Carcinoma

Aim. To investigate the expression of miR-940 in the hepatocellular carcinoma (HCC) and its impact on function and biological mechanism in the HCC cells. Methods. Quantitative RT-PCR analysis was used to quantify miR-940 expression in 46 cases of tissues and cells. Transfection of HCC cell lines was performed by miR-940 mimics; the abilities of invasion and migration were assessed through Trans...
